**Hydrogel-CXCL9 mRNA vaccine** is a preclinical, subcutaneously administered glioblastoma immunotherapy developed at the University of Florida. Also called the hydrogel-chemokine-mRNA vaccine, it co-formulates total tumor-derived mRNA in lipid nanoparticles with the chemokine CXCL9 in a polyethylene glycol hydrogel. The CXCL9-containing depot recruits dendritic cells, natural killer cells, and T cells; recruited antigen-presenting cells take up and express the tumor mRNA payload, promoting tumor-antigen-specific CD8 T-cell responses. In syngeneic murine glioma models, subcutaneous administration produced immune-cell recruitment, tumor-microenvironment reprogramming, and survival benefit; activity was associated with intratumoral CCL4 induction and CD4 T-cell-dependent signaling.
