Hydrolyzed walnut leaf extract is a plant-derived preparation obtained by the hydrolysis of leaves from the Juglans regia (walnut) tree. Walnut leaves are rich in bioactive compounds, including phenolic acids (such as caffeoylquinic and p-coumaroylquinic acids), flavonoids (notably quercetin derivatives), alkaloids, saponins, tannins, and juglone—a naphthoquinone compound that is present as a glycoside precursor in fresh leaves and released upon hydrolysis[6][7]. The extract exhibits strong antioxidant activity due to its high content of polyphenols and flavonoids[8]. Pharmacological studies suggest potential anti-inflammatory, antimutagenic, antiproliferative, antimicrobial, hypoglycemic (via inhibition of protein tyrosine phosphatase 1B and enhancement of glucose uptake), lipid-lowering, and skin-conditioning effects[3][4][6][7]. Traditional uses include topical application for burns or skin irritations and oral use for metabolic syndrome-related conditions such as diabetes or dyslipidemia. The mechanism of action is primarily attributed to antioxidant activity—scavenging reactive oxygen/nitrogen species—and modulation of inflammatory pathways; juglone may also contribute via additional mechanisms such as TLR4/NF-κB pathway suppression[3][7].
