A combination therapy consisting of hydroxychloroquine (HCQ) and vorinostat (VOR). Hydroxychloroquine functions as an autophagy inhibitor while vorinostat is a histone deacetylase (HDAC) inhibitor. This combination has been investigated primarily for metastatic colorectal cancer (mCRC) and HIV. The dual mechanism of action leads to autophagy inhibition, accumulation of ubiquitinated proteins, subsequent tumor cell apoptosis, and improved anti-tumor immunity.
