Overview

Hydroxyl acetylated curcumin (HAC) is a chemically modified derivative of curcumin, where the hydroxyl groups of curcumin are acetylated to increase molecular stability and potentially enhance therapeutic efficacy. HAC has primarily been investigated as a sonosensitizer in sonodynamic therapy (SDT), a technique that uses low-intensity ultrasound to activate certain compounds and induce cytotoxic effects, particularly reactive oxygen species (ROS) generation in target cells. In preclinical studies, HAC-SDT has been shown to induce apoptosis and autophagy in THP-1 derived macrophages, via mitochondrial membrane disruption, release of cytochrome c, and activation of the caspase cascade, as well as modulation of the PI3K/AKT/mTOR pathway. This suggests potential for HAC in anti-atherosclerotic strategies and other applications where targeted induction of cell death or autophagy is beneficial[1][4][7].
