Drug intelligence / Profile preview
hydroxyl-chalcone (also known as DK02) is a synthetic hydroxyl-chalcone derivative being investigated as a multi-target-directed ligand for the treatment of Alzheimer's disease. Designed with structure-guided insights, the molecule features hydroxyl groups and a furan moiety to enhance blood-brain barrier permeability, antioxidant properties, and acetylcholinesterase (AChE) inhibition. In preclinical zebrafish models of scopolamine-induced dementia, hydroxyl-chalcone demonstrated potent AChE inhibition, reduced oxidative stress, decreased amyloid-beta plaque burden, and mitigated tau phosphorylation, leading to significant improvements in memory retention and cognitive function.
