Hydroxymatairesinol lignan (commonly abbreviated as 7-HMR or simply HMR) is a naturally occurring plant lignan found primarily in the knots of Norway spruce (Picea abies) and in various whole grains. It is a precursor to the mammalian enterolignan enterolactone. Hydroxymatairesinol exhibits several biological activities including anti-inflammatory, antioxidant, weak estrogenic/anti-estrogenic effects, and potential anticancer properties. Mechanistically, it acts as a phytoestrogen with much weaker activity than estradiol and can modulate hormone metabolism by inhibiting enzymes such as aromatase and 5α-reductase. Preclinical studies have shown neuroprotective effects in models of Parkinson’s disease by reducing neuroinflammation and slowing dopaminergic degeneration[2][3][5]. Clinical studies indicate that supplementation with hydroxymatairesinol can significantly reduce hot flashes in postmenopausal women[6][8], improve metabolic parameters such as body weight and fat accumulation[9], and may support cardiovascular health. The compound is rapidly absorbed orally and metabolized to enterolactone in humans.
