Overview

HYG-102 is a "soft" estrogenic small molecule originally developed by Hygeia Therapeutics (later acquired by RestorGenex) for the treatment of vulvar and vaginal atrophy (VVA) and age-related skin fragility. Licensed from Yale University, HYG-102 is a biodegradable estradiol derivative, specifically a 15-alpha-substituted estradiol carboxylic acid ester. It is designed to act as a potent estrogen receptor agonist at the site of application while undergoing rapid enzymatic hydrolysis into inactive metabolites upon entering the systemic circulation. This "soft drug" approach is intended to provide the therapeutic benefits of topical estrogen therapy while minimizing systemic exposure and the associated risks of traditional estrogen replacement, such as endometrial or breast stimulation.
