HYNIC-cRGDfk-NPY is a novel heterobivalent dual-receptor-targeting peptide conjugate designed for the imaging of breast tumors. It consists of a cyclic RGD peptide sequence (cRGDfk) that targets integrin αvβ3 (a marker of tumor angiogenesis) and a neuropeptide Y (NPY) analog that targets neuropeptide Y receptors (specifically Y1R, which is overexpressed in approximately 90% of human breast tumors). The peptide is conjugated to hydrazinonicotinamide (HYNIC) to facilitate radiolabeling with technetium-99m (99mTc) for single-photon emission computed tomography (SPECT) imaging. Preclinical studies in breast cancer mouse models (MCF-7 and MDA-MB-231) have demonstrated rapid blood clearance, renal excretion, and high tumor uptake, highlighting its potential as a diagnostic imaging agent.
