Overview

HYP-6589 + Tagrisso is an investigational combination regimen pairing HYP-6589 (also known as HY-0006) with osimertinib (marketed as Tagrisso). HYP-6589 is an oral, highly selective small-molecule inhibitor of Son of sevenless homolog 1 (SOS1), developed by Sichuan Huiyu Pharmaceutical and its subsidiary Sichuan Huiyu Seacross Pharma Technology. SOS1 acts as a guanine nucleotide exchange factor for RAS; by binding to its catalytic region, HYP-6589 reduces KRAS-GTP activation and blocks downstream RAS-RAF-MAPK and RAS-PI3K-AKT-mTOR signaling pathways. Osimertinib is an approved, oral, third-generation EGFR tyrosine kinase inhibitor. The combination is designed to target advanced non-small cell lung cancer (NSCLC) positive for driver gene mutations, aiming to overcome resistance and achieve synergistic anti-tumor efficacy. It is currently being evaluated in a Phase I/II clinical trial (NCT06712680) in China.
