hYP218 CAR-T is an autologous chimeric antigen receptor (CAR) T-cell therapy targeting a membrane-proximal epitope of mesothelin. Developed by researchers at the National Cancer Institute (NCI), this therapy utilizes a humanized antibody fragment (hYP218) that binds to a region of mesothelin closer to the cell membrane than traditional anti-mesothelin agents like SS1. In preclinical studies, hYP218 CAR-T cells demonstrated superior anti-tumor activity, increased cytokine secretion (IL-2, TNFα, IFNγ), and enhanced tumor infiltration compared to distal-binding CAR-T cells. It is currently being investigated in clinical trials for the treatment of mesothelin-expressing solid tumors, including mesothelioma, ovarian cancer, and pancreatic cancer.
