hYP7 CAR-T cells are an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target Glypican-3 (GPC3), a cell-surface oncofetal protein that is highly expressed in hepatocellular carcinoma (HCC) and other solid tumors, while being virtually absent in healthy adult tissues. The therapy utilizes a humanized version of the YP7 single-chain variable fragment (scFv), a high-affinity binder that recognizes a specific epitope on the C-terminal domain of GPC3. By genetically engineering a patient's own T-cells to express this receptor, the cells are redirected to recognize and eliminate GPC3-positive malignant cells. Developed primarily by researchers at the National Cancer Institute, hYP7 CAR-T cells are being evaluated for their safety and efficacy in treating advanced liver cancer, often incorporating costimulatory domains like CD28 or 4-1BB to enhance T-cell persistence and anti-tumor activity within the immunosuppressive tumor microenvironment.
