Overview

hYP7-hYP7 γδAbTCR-T cells are an experimental adoptive cell therapy designed to treat hepatocellular carcinoma (HCC). This therapy utilizes a modular antibody-based γ/δ T-cell receptor (γδAbTCR) platform that grafts the specificity of the hYP7 anti-GPC3 antibody onto a γδ T-cell receptor framework. By using a bivalent hYP7-hYP7 configuration, the cells achieve high antigen-binding avidity, which is particularly effective against tumors with low or heterogeneous Glypican-3 (GPC3) expression. The construct couples antibody-mediated recognition with coordinated TCR-CD3 and CD30 signaling, leading to potent activation of NF-κB and NFAT pathways and inducing rapid caspase-mediated apoptosis in GPC3-positive tumor cells. This architecture is specifically designed to overcome the limitations of antigen mutation and heterogeneity commonly found in advanced liver cancer.
