HyPEP-body is a modular hybrid molecular platform designed for targeted cancer therapy, combining the high tumor penetration of peptides with the long plasma half-life of antibodies. The platform consists of a bispecific peptide pharmacophore labeled with a hapten (such as cotinine) that binds to an anti-hapten antibody through a high-affinity antigen-antibody interaction. In a proof-of-concept study, a HyPEP-body was constructed to simultaneously target Extra Domain B (EDB) of fibronectin and Vascular Endothelial Growth Factor (VEGF). The antibody component extends the circulation time of the peptides, which are subsequently released to penetrate solid tumor tissues more effectively than full-sized antibodies. Preclinical studies in glioblastoma xenograft models demonstrated significant tumor growth inhibition.
