Overview

Hyperactive PASylated DNase I is an investigational biologic therapeutic being developed by XL-protein in collaboration with Rentschler Biopharma. The drug consists of a hyperactive variant of recombinant human deoxyribonuclease I (DNase I) that has been modified using XL-protein's proprietary PASylation technology. This technology involves the genetic fusion of the enzyme with a defined polypeptide sequence of proline, alanine, and serine (PAS), which significantly increases the enzyme's systemic half-life and enhances its catalytic activity compared to the wild-type enzyme. The therapeutic is designed to target and degrade extracellular DNA, including neutrophil extracellular traps (NETs), which are known to drive inflammation and increase mucus viscosity in various pathological conditions. It is currently in preclinical development for the treatment of cystic fibrosis, autoimmune diseases, and other chronic inflammatory disorders.
