Overview

The "HyperAcute vaccine" refers to a family of allogeneic whole-cell cancer vaccines developed by NewLink Genetics. These vaccines are genetically engineered tumor cell lines that express the murine alpha-galactosyl (alpha-gal) epitope on their surface. When injected into humans, these cells trigger a hyperacute rejection response mediated by pre-existing human anti-alpha-gal antibodies. This immune reaction leads to rapid destruction of the injected cells and stimulates an anti-tumor immune response against similar antigens present on the patient's own tumor cells. Each "HyperAcute" variant is tailored for a specific cancer type (e.g., pancreas, lung, prostate). The most well-known member is algenpantucel-L for pancreatic cancer[4][7][9].
