HypoSti.CAR-HER2 T cells are a novel form of chimeric antigen receptor (CAR) T cell therapy engineered to target human epidermal growth factor receptor 2 (HER2), which is overexpressed in various advanced solid tumors. This product utilizes a hypoxia-stimulated CAR expression system (HypoSti.CAR), designed to enhance the expansion and survival of CAR-T cells within the hypoxic tumor microenvironment—a common feature of solid tumors that often limits the efficacy of conventional CAR-T therapies. By restricting robust CAR expression to low-oxygen conditions, this approach aims to improve anti-tumor activity while reducing off-tumor toxicity. The safety, feasibility, and preliminary efficacy of HypoSti.CAR-HER2 T cells are being evaluated in phase 1/2 clinical trials for patients with HER2-positive advanced solid tumors[1][3][4][9].
