Hypotonic saline is a sodium chloride solution with an osmolarity lower than that of physiological saline (0.9% NaCl). In clinical research, it is frequently utilized as a control or placebo to evaluate the efficacy of therapeutic agents in ocular and respiratory diseases. For example, it served as the comparator arm in the Phase 3 GOBI and MOJAVE trials for Miebo (perfluorohexyloctane) in patients with dry eye disease (DED) and meibomian gland dysfunction (MGD). In respiratory medicine, hypotonic saline (e.g., 0.12% NaCl) is used as a control to study mucociliary clearance and lung function in conditions such as chronic obstructive pulmonary disease (COPD) and cystic fibrosis, where it typically lacks the therapeutic mucus-clearing properties of hypertonic saline.
