Overview

A therapeutic intervention involving controlled exposure to low-oxygen air (hypoxia) to treat Parkinson's disease. Developed by Radboud University Medical Center, this intervention utilizes normobaric hypoxia delivered at various fractions of inspired oxygen (FiO2), including continuous hypoxia (FiO2 0.163 and 0.127) and intermittent hypoxia protocols. The therapy works by activating the Hypoxia Inducible Factor 1 (HIF-1) pathway, which in turn stimulates tyrosine hydroxylase (TH), the rate-limiting enzyme in dopamine production. This process is intended to increase dopamine release from the substantia nigra and provide neuroprotective effects through hypoxic preconditioning. The treatment is currently being evaluated in the TALISMAN Phase 1 clinical trial (NCT05214287).
