HZ-A-018

HZ-A-018 is a highly selective, potent, covalent small-molecule inhibitor of Bruton tyrosine kinase (BTK). It is designed to block the B-cell receptor (BCR) and Toll-like receptor signaling pathways, thereby inhibiting the proliferation of B cells and promoting apoptosis. Notably, it demonstrates high permeability across the blood-brain barrier, making it particularly suitable for central nervous system (CNS) lymphomas. Preclinical and early clinical studies indicate that HZ-A-018 has favorable pharmacokinetics and safety profiles. It has shown efficacy as monotherapy in relapsed/refractory primary CNS lymphoma (R/R PCNSL), with ongoing trials also exploring its use in other B-cell malignancies and immune-mediated diseases such as chronic/persistent adult primary immune thrombocytopenia, multiple sclerosis, and neuromyelitis optica spectrum disorders.
