Overview

HZ-A-018 is a highly selective, potent, covalent small-molecule inhibitor of Bruton tyrosine kinase (BTK). It is designed to block the B-cell receptor (BCR) and Toll-like receptor signaling pathways, thereby inhibiting the proliferation of B cells and promoting apoptosis. Notably, it demonstrates high permeability across the blood-brain barrier, making it particularly suitable for central nervous system (CNS) lymphomas. Preclinical and early clinical studies indicate that HZ-A-018 has favorable pharmacokinetics and safety profiles. It has shown efficacy as monotherapy in relapsed/refractory primary CNS lymphoma (R/R PCNSL), with ongoing trials also exploring its use in other B-cell malignancies and immune-mediated diseases such as chronic/persistent adult primary immune thrombocytopenia, multiple sclerosis, and neuromyelitis optica spectrum disorders. The drug was developed by Hangzhou HealZen Therapeutics Co., Ltd (also referenced as Hangzhou Hezheng Pharmaceutical or Hangzhou Hertz Pharmaceutical), an innovative Chinese pharmaceutical company focused on targeted therapies for hematological tumors and CNS diseases[2][3][5][7].
