HZ-H08905 is a novel, potent, oral small molecule dual inhibitor targeting casein kinase 1 epsilon (CK1ε) and phosphatidylinositol 3-kinase delta (PI3Kδ). It was independently developed by Hangzhou HealZen Therapeutics. The drug is designed to treat hematologic malignancies by inhibiting both CK1ε and PI3Kδ pathways, which are implicated in the pathogenesis of lymphoma. Dual inhibition leads to direct induction of apoptosis and inhibition of tumor cell proliferation in lymphoma cells. Additionally, CK1ε inhibition may regulate immunosuppression via the Wnt/β-Catenin signaling pathway by maintaining Treg survival. Preclinical studies demonstrated strong anti-tumor activity in B-cell lymphoma models and suggested a differentiated safety profile compared to other PI3Kδ inhibitors. Clinical trials have shown promising efficacy and good tolerability for relapsed/refractory peripheral T-cell lymphoma (PTCL) and B-cell lymphomas[2][3][4][5][6].
