**HzMUC1-MMAE** is an investigational antibody-drug conjugate (ADC) consisting of a humanized monoclonal antibody targeting the cancer-associated glycoprotein Mucin 1 (MUC1), conjugated via a valine-citrulline dipeptide linker to the cytotoxic payload monomethyl auristatin E (MMAE). The antibody (HzMUC1) is specific to the region between extracellular subunits MUC1-N and MUC1-C. Upon binding to MUC1-expressing tumor cells, HzMUC1-MMAE is internalized, where intracellular release of MMAE disrupts microtubule polymerization, resulting in G2/M cell cycle arrest and apoptosis. Preclinical studies have demonstrated potent anti-tumor activity in MUC1-positive pancreatic cancer models and in trastuzumab-resistant HER2-positive breast cancer models[1][2][5].
