IκBα-SR mRNA is a therapeutic messenger RNA encoding a super-repressor (SR) mutant of the Inhibitor of nuclear factor kappa B alpha (IκBα). This mutant typically features serine-to-alanine substitutions at positions 32 and 36, which prevent its phosphorylation by the IκB kinase (IKK) complex. Consequently, the protein is not targeted for proteasomal degradation and remains stably bound to the NF-κB transcription factor complex in the cytoplasm. By preventing the nuclear translocation of NF-κB, IκBα-SR mRNA effectively blocks the transcription of a wide array of pro-inflammatory cytokines and mediators. Developed primarily by Ethris using their SNIMRNA (Stabilized Non-Immunogenic mRNA) platform, this candidate is being investigated for the treatment of acute and chronic inflammatory conditions, including acute lung injury (ALI), sepsis, and COVID-19-related pulmonary inflammation.
