Overview

I-131-CLR1404 + dexamethasone is an investigational combination therapy under clinical evaluation for relapsed or refractory multiple myeloma. I-131-CLR1404 is a small-molecule, cancer-targeted radiopharmaceutical composed of a proprietary phospholipid ether (PLE) analog, which selectively accumulates in malignant cells, covalently labeled with iodine-131, a cytotoxic radioisotope. This agent delivers targeted intracellular ionizing radiation that kills cancer cells, including cancer stem cells, by exploiting their unique lipid metabolism and membrane composition. Dexamethasone, a synthetic glucocorticoid, is used concurrently for its established anti-myeloma and anti-inflammatory effects. The combination aims to enhance efficacy in treating patients who have failed standard therapies. The drug candidate was granted orphan drug designation in multiple myeloma and is being developed by Cellectar Biosciences[1][3][5].
