IACS-8779 is a small-molecule agonist of the Stimulator of Interferon Genes (STING) pathway, developed by the Institute for Applied Cancer Science (IACS) at MD Anderson Cancer Center. It is designed to trigger a robust innate antitumor immune response, particularly in immunologically "cold" tumors like glioblastoma that typically lack significant immune cell infiltration. By activating STING, the drug promotes the production of Type I interferons and other pro-inflammatory cytokines, which in turn enhances the recruitment and activation of tumor-specific T cells within the tumor microenvironment. IACS-8779 has been evaluated in Phase I clinical trials in canine patients with spontaneously arising glioblastoma, where it demonstrated dose-dependent radiographic responses and a manageable safety profile when administered intratumorally via stereotactic navigation.
