IAM-C1 is a first-in-class small molecule inhibitor developed by Iambic Therapeutics. It is designed to selectively inhibit cyclin-dependent kinases CDK2 and CDK4, two cell-cycle kinases frequently dysregulated in various cancers. Unlike currently approved CDK4/6 inhibitors, IAM-C1 preserves the activity of other closely related kinases such as CDK1, CDK6, and CDK9. This selectivity aims to provide expanded therapeutic benefits and a more favorable safety profile by reducing dose-limiting toxicities associated with non-CDK2/4 inhibition. The drug is being developed primarily for hormone receptor positive (HR+), HER2 negative breast cancer, particularly to address resistance seen with existing therapies[1][3][7][9].
