Overview

IAMA-004 is a long-acting tri-specific antibody designed to target human epidermal growth factor receptor 2 (HER2) as its primary target, alongside CD47 and CD16a (FcγRIIIa) as effector cell engagers. Developed by Nanjing JSIAMA Biopharmaceuticals, the molecule is engineered to engage both macrophages (via CD47) and natural killer (NK) cells (via CD16a) to promote innate immune-mediated tumor cell killing. CD47 also serves as a co-target on HER2/CD47 double-positive tumor cells. In preclinical studies, IAMA-004 has demonstrated potent antibody-dependent cellular cytotoxicity (ADCC) and tumor cell killing in patient-derived ex-vivo fluids, such as malignant pleural effusion and malignant ascites, particularly when combined with the Fc-cytokine fusion protein IAMA-005.
