Drug intelligence / Profile preview
IAR-125 is a novel bispecific monoclonal antibody designed to simultaneously target and inhibit tumor necrosis factor-like ligand 1A (TL1A) and interleukin-23 subunit alpha (IL-23p19). Developed by Innovent Biologics, IAR-125 utilizes a 2+2 geometry design to prevent the formation of large immune complexes, thereby reducing the risk of anti-drug antibodies (ADA) typically associated with TNF family members. The drug is being developed for the treatment of inflammatory bowel disease (IBD). In preclinical models, IAR-125 demonstrated superior blocking efficacy of both TL1A and IL-23 compared to benchmark molecules, leading to broader and deeper inhibition of disease-driving inflammation. It also exhibited a favorable safety and pharmacokinetic profile in non-human primates, supporting its potential for subcutaneous administration with less frequent dosing.
