Overview

IARS siRNA is a small interfering RNA (siRNA) designed to target and silence the expression of isoleucyl-tRNA synthetase (IARS), also known as IARS1. IARS is an enzyme primarily responsible for ligating isoleucine to its cognate tRNA, but it has also been implicated in non-canonical inflammatory signaling pathways. In the context of dermatological research, particularly psoriasis, IARS siRNA has been shown to inhibit the Jak2/STAT3/CXCL16 signaling pathway in human epidermal keratinocytes. By reducing IARS protein levels, the siRNA suppresses the production of pro-inflammatory cytokines and chemokines, including IL-6, IL-1β, CCL5, and CXCL16, thereby ameliorating psoriasis-like skin lesions in experimental models. This approach represents a potential therapeutic strategy for inflammatory skin diseases by modulating the aminoacyl-tRNA synthetase-mediated inflammatory axis.
