IASO 206 is an investigational in vivo CAR-T cell therapy developed by IASO Bio for the treatment of multiple myeloma. Unlike traditional CAR-T therapies that require complex ex vivo manufacturing, IASO 206 utilizes a third-generation lentiviral vector platform to generate CAR-T cells directly within the patient's body. The vector is engineered with a mutated cocal glycoprotein to disrupt native LDL-R binding and a high-affinity CD7 binder to specifically target T cells. It delivers a transgene encoding a chimeric antigen receptor (CAR) that targets B-cell maturation antigen (BCMA) using a humanized VHH nanobody. To enhance the lifespan and persistence of the generated CAR-T cells, the platform incorporates CD47 expression and MHC I gene knockout to reduce macrophage phagocytosis and T-cell mediated elimination. Preclinical studies have demonstrated that a single intravenous administration of IASO 206 can lead to significant tumor regression in multiple myeloma xenograft models.
