Overview

IAV-OX40L is a genetically engineered **chimeric oncolytic influenza A virus** that has been modified to express the **OX40 ligand (OX40L) gene** within its nonstructural (NS1) protein gene. This modification enables the virus to deliver OX40L to tumor microenvironments upon infection. The mechanism of action leverages both the cytolytic properties of the influenza virus and the immune-activating effects of OX40L; the latter augments antitumor immunity by **stimulating Th1-dominant immune responses**, increasing the activation and proliferation of **CD4+ and CD8+ T cells**, and enhancing the secretion of interferon-γ and interleukin-2. In preclinical models of hepatocellular carcinoma (HCC), IAV-OX40L demonstrated selective cytotoxicity for tumor cells and significant tumor inhibition in mice with minimal impact on normal tissues. The therapy is being investigated as a novel virotherapy platform for treating cancers, specifically liver cancer[5].
