Overview

IAXO-102 is a synthetic small-molecule glycolipid and benzylammonium lipid that acts as a Toll-like receptor 4 (TLR4) and CD14 antagonist. Developed by Innaxon, it is a lipid A analogue designed to modulate TLR4 signaling pathways. Mechanistically, IAXO-102 negatively regulates TLR4 signaling by inhibiting the phosphorylation of MAPK and p65 NF-κB, thereby downregulating the expression of TLR4-dependent proinflammatory proteins. It has demonstrated preclinical efficacy in various inflammatory and vascular conditions, including abdominal aortic aneurysm (AAA), neuropathic pain, atherosclerosis, and acute organ failure. The compound is also utilized as a research tool to investigate CD14-dependent and TLR4-independent pathways in sterile inflammation.
