Drug intelligence / Profile preview
IB3D is a sustained-release intravitreal implant containing Kamuvudine-8 (K8), a small molecule inhibitor of the NLRP3 inflammasome. Developed by Inflammasome Therapeutics, IB3D is designed for the treatment of geographic atrophy (GA) secondary to age-related macular degeneration (AMD) and diabetic macular edema (DME). Kamuvudines represent a novel class of modified nucleosides that lack the 5'-hydroxyl group, which prevents their phosphorylation and subsequent incorporation into DNA, thereby avoiding the mitochondrial and systemic toxicities associated with traditional nucleoside reverse transcriptase inhibitors (NRTIs). By specifically inhibiting the NLRP3 inflammasome, IB3D targets the chronic inflammatory response and subsequent cell death of retinal pigment epithelium (RPE) cells, which are central to the progression of GA. The implant is engineered to provide a steady release of the active drug over a period of several months, reducing the burden of frequent intravitreal injections.
