IB4000 is a small molecule drug candidate developed by IntraBio Ltd. for the treatment of various lysosomal storage disorders (LSDs), including Gaucher disease, Tay-Sachs disease, and Sandhoff disease. It functions as a glucosylceramide synthase (GCS) inhibitor, which is a form of substrate reduction therapy (SRT). By inhibiting the enzyme ceramide glucosyltransferase (encoded by the UGCG gene), IB4000 reduces the biosynthesis of glucosylceramide, the precursor for more complex glycosphingolipids. This reduction helps prevent the pathological accumulation of these lipids in cells, particularly in the central nervous system and visceral organs. IB4000 is designed to be orally bioavailable and blood-brain barrier penetrant, aiming to address the significant unmet need for therapies that can treat the neurological manifestations of LSDs.
