Overview

**iBCL11B-hiPSC-derived NK cells** are engineered natural killer (NK) cells derived from human induced pluripotent stem cells (hiPSCs) genetically modified to overexpress BCL11B, a transcription factor that modulates hemogenic endothelium-derived multipotent progenitors. These cells exhibit mature NK-specific surface markers such as CD56, NKG2D, NKp44, and NKp46, and demonstrate potent cytotoxicity against cancer cells, including K562 leukemia lines, through mechanisms like granule-mediated lysis, cytokine production (e.g., IFN-γ, TNFα), and death receptor pathways. Developed as an off-the-shelf cell therapy platform, they offer advantages in scalability, genetic engineering uniformity, and reduced risk of graft-versus-host disease compared to primary NK cells, with potential applications in cancer immunotherapy.[3][1][2]
