IBE-814 IVT is a novel, fully degradable intravitreal implant designed to deliver a sustained, low dose of dexamethasone directly to the retina for at least six months. The implant is unique in that it does not use any polymeric carriers or excipients; instead, it consists solely of a dexamethasone prodrug (IBE-814) engineered using Ripple Therapeutics' Epidel technology platform. This allows for zero-order drug release kinetics via surface erosion, resulting in consistent and prolonged delivery with a much smaller implant size that can be administered through a 30-gauge needle. The primary indications under investigation are diabetic macular edema (DME) and retinal vein occlusion (RVO), with the goal of reducing treatment frequency and minimizing steroid-related adverse events compared to existing therapies. Clinical studies have shown sustained reductions in central subfield thickness (CST), improved or stable best-corrected visual acuity (BCVA), and reduced treatment burden over at least six months[1][3][8][10].
