Ibentatug is a humanized IgG1 kappa monoclonal antibody that selectively targets cancer-specific plectin 1 (CSP1), a cell-surface form of plectin (PLEC) aberrantly expressed on a wide range of solid tumors and associated with aggressive disease and poor prognosis.[1][3][13] By binding CSP1, ibentatug induces cytotoxicity through activation of cellular stress pathways and downregulation of survival and proliferation signaling cascades including SRC, PRAS40, STAT3, AKT, and ERK1/2, resulting in apoptosis and inhibition of tumor growth in preclinical models.[3] Developed by ZielBio as its lead oncology asset, ibentatug is being evaluated in a first‑in‑human phase 1/2 trial in advanced solid tumors, with focus on cholangiocarcinoma, pancreatic (including pancreatic ductal adenocarcinoma) and ovarian cancers, and has received FDA Orphan Drug Designation for cholangiocarcinoma and pancreatic cancer.[1][3][6][11]
