Overview

**Iberdomide** is an oral small-molecule cereblon E3 ligase modulator developed by Celgene and later Bristol Myers Squibb. It acts as a molecular glue that binds cereblon and promotes ubiquitination and proteasomal degradation of the transcription factors Ikaros and Aiolos, encoded by **IKZF1** and **IKZF3**, respectively. This results in broad immunomodulatory effects including depletion of B cells and plasmacytoid dendritic cells, reduction of type I interferon pathway activity and autoantibody production, and increases in interleukin-2 and regulatory T cells. Publicly documented development has focused primarily on **systemic lupus erythematosus**, including phase 2 studies showing SRI-4 benefit at 0.45 mg daily and biomarker-enriched activity in patients with high type I interferon or Aiolos signatures; it has also been studied for cutaneous manifestations of lupus and in oncology combinations such as relapsed or refractory multiple myeloma.
