IBI-307 is an investigational antibody-drug conjugate (ADC) developed by Innovent Biologics that targets Claudin 18.2 (CLDN18.2), a tight junction protein frequently overexpressed in various solid tumors, including gastric, gastroesophageal junction, and pancreatic cancers. The drug consists of a humanized monoclonal antibody specific to CLDN18.2 conjugated to a potent cytotoxic payload via a linker. Upon binding to CLDN18.2 on the surface of tumor cells, the ADC is internalized, and the payload is released, leading to cell cycle arrest and apoptosis. IBI-307 is designed to provide targeted delivery of chemotherapy, thereby enhancing efficacy while minimizing systemic toxicity. It has been evaluated in Phase 1 clinical trials for patients with advanced solid tumors expressing CLDN18.2.
