IBI-334 is a bispecific antibody that targets both B7-H3 (CD276) and epidermal growth factor receptor (EGFR). It is designed to block EGFR signaling while simultaneously engaging B7-H3, which is often co-expressed with EGFR in multiple solid tumors. The antibody is afucosylated to enhance antibody-dependent cellular cytotoxicity (ADCC), thereby increasing its anti-tumor efficacy. Preclinical studies have shown that IBI-334 provides more potent inhibition of EGFR signaling compared to existing monoclonal antibodies and other bispecific antibodies targeting similar pathways. Developed by Innovent Biologics, IBI-334 has demonstrated a favorable safety profile in animal models and shows broad application potential across various EGFR-driven solid tumors, including colorectal, breast, gastric, gastrointestinal, head and neck, lung, pancreatic cancers, and other advanced malignant solid neoplasms[1][2][5][6][8].
