IBI110 is a fully human IgG4κ recombinant monoclonal antibody that targets lymphocyte activation gene 3 protein (LAG-3; CD223), a co-inhibitory immune checkpoint receptor expressed on activated T cells. By binding to LAG-3, IBI110 inhibits its interaction with MHC class II molecules and other ligands, thereby blocking the negative regulatory signal that suppresses T cell activation. This results in enhanced T cell proliferation and cytokine production, promoting anti-tumor immune responses. The drug is being developed primarily as an immunotherapy for various cancers and has been studied both as monotherapy and in combination with other agents such as sintilimab (an anti-PD-1 antibody) and chemotherapy. It is under development by Innovent Biologics for indications including squamous non-small cell lung cancer (sqNSCLC), gastric cancer, hepatocellular carcinoma (HCC), small cell lung cancer, lymphoma, solid tumors, alveolar soft part sarcoma, esophageal squamous cell carcinoma, adenocarcinoma of the gastroesophageal junction, among others[2][9][10].
