Overview

IBI3001 is a novel, potentially first-in-class bispecific antibody-drug conjugate (ADC) targeting both B7-H3 (CD276) and EGFR. Developed using the SYNtecanE platform for site-specific glycan conjugation, it is designed to enhance anti-tumor efficacy while reducing on-target toxicities associated with EGFR inhibition. The drug exerts multiple anti-tumor mechanisms of action including enhanced blockade of EGFR signaling, dual-targeted payload internalization and cytotoxicity via both B7-H3 and EGFR, as well as potent bystander killing effects. Preclinical studies have demonstrated strong in vitro and in vivo efficacy across various solid tumors with favorable pharmacokinetic and safety profiles. IBI3001 is being developed by Innovent Biologics for the treatment of solid tumors and is currently in phase 1 clinical trials[1][2][4][5][7].
