IBI3016 is a small interfering RNA (siRNA) drug candidate developed to target angiotensinogen (AGT) for the treatment of hypertension. By silencing the AGT gene, IBI3016 reduces serum levels of AGT and associated biomarkers such as angiotensin I and II, leading to significant and sustained reductions in blood pressure. Preclinical studies in hypertensive cynomolgus monkeys demonstrated marked efficacy without notable safety concerns like hypotension. The drug utilizes SanegeneBio’s next-generation proprietary siRNA technology platform, which enhances potency and durability while maintaining favorable safety and tolerability profiles. As of August 2024, IBI3016 is being evaluated in a Phase 1 first-in-human clinical trial (NCT06501586) involving healthy volunteers and patients with mild hypertension. The program is co-developed by SanegeneBio and Innovent Biologics under a strategic partnership established in December 2023; Innovent holds an exclusive option for future development, manufacturing, and commercialization rights[1][2][4][5][6][8].
