IBI3017 is an innovative, first-in-class bispecific antibody-drug conjugate (bsADC) developed by Innovent Biologics. It simultaneously targets Trophoblast cell surface antigen 2 (Trop2) and B7 homolog 3 (B7H3), two tumor-associated antigens frequently co-expressed in solid tumors such as non-small cell lung cancer (NSCLC), prostate adenocarcinoma, and breast cancer. The molecule consists of a bispecific antibody conjugated to a novel topoisomerase I inhibitor payload, NT1, with a drug-to-antibody ratio (DAR) of 8. A key design feature of IBI3017 is its reduced binding affinity for Trop2 relative to B7H3, which is intended to enhance selectivity for tumors co-expressing both targets while minimizing on-target/off-tumor toxicities, such as stomatitis and mucositis, commonly associated with Trop2-targeted ADCs. Preclinical data indicate superior anti-tumor activity and a favorable safety profile compared to parental monospecific ADCs.
