Overview

IBI3019 is a novel tri-specific antibody developed for the treatment of colorectal cancer. It targets three distinct antigens: epidermal growth factor receptor (EGFR), cadherin-17 (CDH17), and Fc gamma receptor IIIa (CD16A). By simultaneously binding to EGFR and CDH17, which are overexpressed on tumor cells, and engaging CD16A on immune effector cells, IBI3019 is designed to enhance tumor-specific cytotoxicity while reducing off-tumor toxicities such as skin reactions commonly associated with EGFR-targeted therapies. Preclinical studies have demonstrated that IBI3019 exhibits superior anti-tumor efficacy compared to existing agents like cetuximab and amivantamab, along with a favorable safety profile in animal models[1][3][4].
