IBI3031 is an investigational, fully human monoclonal antibody targeting the insulin-like growth factor 1 receptor (IGF-1R), developed by Innovent Biologics for the treatment of thyroid eye disease (TED). TED is an autoimmune condition characterized by inflammation and expansion of orbital tissues, leading to symptoms such as proptosis (eye bulging), diplopia (double vision), and orbital pain. The IGF-1R pathway plays a critical role in the activation of orbital fibroblasts, which drives the pathogenesis of the disease. IBI3031 acts as an antagonist, binding to IGF-1R and blocking its activation by ligands such as IGF-1 and IGF-2. While Innovent's lead IGF-1R candidate, IBI311, is in late-stage development for intravenous administration, IBI3031 is a next-generation candidate currently in Phase 1 clinical trials (e.g., CTR20261962) to assess its safety, tolerability, and pharmacokinetics, potentially offering an optimized profile or alternative administration route such as subcutaneous injection.
