Overview

IBI3033 is a long-acting triple agonist targeting the glucagon-like peptide-1 receptor (GLP-1R), glucose-dependent insulinotropic polypeptide receptor (GIPR), and glucagon receptor (GCGR). Developed by Innovent Biologics, it is designed for the treatment of metabolic disorders, including obesity and type 2 diabetes mellitus. By activating all three receptors, the drug aims to leverage the synergistic effects of GLP-1 and GIP on insulin secretion and appetite suppression, alongside the energy expenditure-increasing properties of glucagon, potentially offering superior weight loss and glycemic control compared to mono- or dual-agonists. As of early 2024, it is in Phase 1 clinical development to evaluate its safety, tolerability, and pharmacokinetics in healthy participants.
