Overview

IBI321 is a bispecific monoclonal antibody designed to simultaneously target and inhibit both programmed cell death protein 1 (PD-1) and T cell immunoreceptor with Ig and ITIM domains (TIGIT). By blocking these two immune checkpoint pathways, IBI321 aims to restore immune function through the activation of T cells, thereby enhancing anti-tumor immune responses. Preclinical studies have shown that IBI321 retains the activity of a PD-1 inhibitor while also suppressing TIGIT signaling, resulting in improved efficacy compared to targeting either pathway alone. The drug was discovered through collaboration between Innovent Biologics and Eli Lilly and Company, with clinical development led by Innovent in China for advanced solid tumors[1][2][3][5].
