IBI323 is a recombinant humanized bispecific monoclonal antibody that targets both lymphocyte activation gene 3 (LAG-3) and programmed death-ligand 1 (PD-L1). It is designed to block the interaction of LAG-3 with MHC-II and PD-L1 with PD-1/CD80, thereby enhancing T cell activation and reversing tumor-mediated immune suppression. Preclinical studies have shown that IBI323 can bridge PD-L1+ cells and LAG-3+ cells, leading to superior immune stimulatory activity compared to parental antibodies. The drug has demonstrated enhanced antitumor effects in humanized mouse models by increasing tumor-specific CD8+ and CD4+ T cell responses. Developed by Innovent Biologics, IBI323 is being investigated primarily for the treatment of solid tumors[2][4][5][7].
