Overview

IBI324 is a recombinant fully humanized bispecific antibody developed by Innovent Biologics. It targets both vascular endothelial growth factor A (VEGF-A) and angiopoietin-2 (Ang-2), acting as an inhibitor of both pathways. The N-terminal domain of IBI324 blocks VEGF-A-mediated signaling, inhibiting the growth and proliferation of vascular endothelial cells, thereby suppressing angiogenesis and reducing vascular permeability and leakage. The C-terminal domain binds Ang-2, preventing its interaction with the Tie-2 receptor, which further stabilizes blood vessels and inhibits inflammation-induced leakage. This dual mechanism aims to provide anti-angiogenic effects, vascular stabilization, and inflammation inhibition for ophthalmic diseases such as diabetic macular edema (DME). Preclinical studies have shown that IBI324 can inhibit neovascularization and reduce vascular leakage, potentially allowing for longer dosing intervals compared to current therapies[1][5][7].
