IBI343 + gemcitabine + albumin-bound paclitaxel is an investigational combination therapy being developed by Innovent Biologics for the treatment of advanced solid tumors, specifically claudin 18.2 (CLDN18.2)-positive gastric adenocarcinoma, gastroesophageal junction adenocarcinoma, and pancreatic ductal adenocarcinoma. IBI343 is a recombinant human anti-CLDN18.2 monoclonal antibody-drug conjugate (ADC) utilizing a topoisomerase I inhibitor payload (exatecan). It targets tumor cells expressing CLDN18.2, leading to internalization and the release of the cytotoxic payload. Gemcitabine is a nucleoside metabolic inhibitor that interferes with DNA synthesis, while albumin-bound paclitaxel (nab-paclitaxel) is a microtubule inhibitor that disrupts mitotic processes. This combination therapy aims to enhance anti-tumor efficacy by combining targeted ADC delivery with systemic chemotherapy in patients with CLDN18.2-expressing malignancies.
